After sitting down on the computer for long periods of time I get this knot or somethign in my upper right back. How can I get rid of this?

Go see somebody. Could either be just bad posture, or you have an imbalance somewhere thats pulling it.
The only way you'll find out is to see a doc or something.

Sit in a chair instead.

Totally didn't notice that, lol.

Improve posture and take breaks from sitting.
If that doesn't work, go see a doc.

Damn...
Danny, you, like so many other Americans, have a shitty posture.
8 years ago I decided to not slouch when sitting, or at anytime really. It takes a couple of months to get there. It'll also cause pain at first. You're working muscles that you haven't been.
My current chair doesn't even have a back on it.

Sit up straight.

unless you have a unatural curve to your spine, sitting up straight will fix the problem.
