I would say not flexed, except when measuring bicep.
When checking proportions, should muscles be flexed or not?

I'm already off of that chart
I dont flex everything, but chest is one thing that will change based on how much air you have in you, so I just go ahead and flex the lats as well as breathe in. At least pick something and stick to it. Calves and quads I dont flex, when I measure my "gut" I just let it hang out which keeps things consistent.
So to measure improvement I would just make sure you pick a method and stick to it. Same with bodyfat calipers, you dont even need to calculate, you can just measure the mm of the skinfold.
